By Sarah Crunkhorn Verified| Nature Verified IN BRIEF 06 May 2022 The mechanism by which the immunotherapeutic target LAG3 — an inhibitory receptor expressed on activated T cells — inhibits T cell function remains unknown. Using T cells isolated from mice, Guy et al. show that LAG3 can inhibit T cell function in the absence of its canonical ligand, MHC class II. Of note, existing LAG3 therapeutics target the LAG3–MHC class II interaction.
By Sarah Crunkhorn Verified| Nature Verified IN BRIEF 06 May 2022 Natural killer (NK) cell-based immunotherapy is an attractive anticancer strategy, but efficacy in clinical trials has been limited. Here, Pan et al. demonstrate that NK cells induce mitochondrial apoptosis of cancer cells, which is essential for NK cell-mediated killing. BH3 profiling — which measures mitochondrial sensitivity to BH3 peptides — showed that NK cells primed cancer cells for mitochondrial apoptosis.
